Q. Is there any at home test that I can do to determine the difference between engine oil and transmission fluid using only one drop of each?

A. I don't know of any test kit available that will do the kind of test you want to do with any amount of fluid. Usually there is enough fluid present to determine oil or transmission fluid simply by color. Transmission fluid is red and oil is from a light honey to a dark honey color.
